BPC-157 and TB-500 blend: composition and analytical characterisation

Composition note for the two-component 30 mg preparation: what each compound is, how the split is recorded, and how one certificate of analysis carries a separate purity and identity line for each.

Composition

The preparation is a single lyophilised powder containing two compounds, BPC-157 and TB-500, with a total peptide mass of 30 mg per vial. The mass of each component has not been confirmed, so no per-compound split is stated; the catalogue record carries the total quantity only until a split is verified against a certificate.

BPC-157 is a synthetic pentadecapeptide of sequence GEPPPGKPADDAGLV, recorded with CAS 137525-51-0, UNII 8ED8NXK95P, PubChem CID 9941957, molecular formula C62H98N16O22 and a molecular weight of 1419.5 g/mol 123. A separate substance record exists for the acetate salt, so the salt form is an identity question in its own right 4.

TB-500 is a trade name applied to two different molecules: the synthetic N-acetylated heptapeptide Ac-Leu-Lys-Lys-Thr-Glu-Thr-Gln, registered under UNII QHK6Z47GTG with CAS 885340-08-9, formula C38H68N10O14 and a molecular weight of 889.0 g/mol, and full-length thymosin β4, the 43-residue peptide given the international non-proprietary name timbetasin 5678. Which molecule is supplied is unconfirmed, and FDA states that the two names are used interchangeably although they are not the same substance 7.

Analytical characterisation

Each compound in a multi-component preparation is identified and quantified separately on the same certificate of analysis. The data model pairs one certificate with one compound and records, per compound: a chromatographic purity figure with the high-performance liquid chromatography method used, and a mass-spectrometric identity line giving the method, the expected and observed masses and whether identity was confirmed, together with the assayed mass and net peptide content. A two-component preparation therefore produces two such rows on one certificate, and neither a combined purity figure nor a combined mass is meaningful across the pair.

Neither compound has a United States Pharmacopeia, National Formulary or European Pharmacopoeia monograph, so no compendial specification or official reference standard can be quoted for either line 37.
