In solution the complex is monomeric with three nitrogen donors (glycine amino N, deprotonated amide N, histidine imidazole N); in the solid state it is dimeric, with the fourth equatorial site occupied by a labile carboxylate oxygen. The lysine side-chain amine takes part in coordination only at alkaline pH.

Research overview

GHK-Cu is the 1:1 copper(II) complex of the tripeptide glycyl-L-histidyl-L-lysine, and it is a separate registry substance from the free tripeptide. The complex is registered as prezatide copper, CAS 89030-95-5, UNII 6BJQ43T1I9, with the INCI name Copper Tripeptide-1; the free peptide is registered as prezatide, CAS 49557-75-7, C14H24N6O4, 340.38 g/mol 12. Two molecular weights are published for the complex: 401.91 g/mol for the neutral representation held by the substance registry and by PubChem CID 10111685, and 402.92 g/mol for the cationic representation on CID 71587328, the record that carries the CAS number and the UNII. The difference is one proton in how the same complex is drawn, not two materials 34. Structural work describes a complex that is monomeric in solution, with three nitrogen donors and a labile fourth equatorial site, and reports a conditional dissociation constant at pH 7.4 of 7.0 plus or minus 1.0 times 10 to the power minus 14 M 56. The free tripeptide glycyl-L-histidyl-L-lysine is a separate record, CAS 49557-75-7, UNII 39TG2H631E, INCI name Tripeptide-1 [gsrs-prezatide][pubchem-73587].
